Çalışmada materyal olarak, 20 adet erişkin (6 aylık ve sağlıklı)bıldırcın pankreası kullanıldı.Makroskobik incelemelerde dorsal, ventral, üçüncü lop ve dalak lopları bulunanbıldırcın pankreasının duodenumun kolları arasına yerleştiği görüldü. Pankreasın ekzokrinkısmını oluşturan asinuslar ve akıtıcı kanallarının histolojik özellikleri belirlendi. Asinuslardabulunan piramidal yapılı salgı hücrelerinin bazalde yer alan çekirdek, granüllü endoplazmikretikulum ve apikalde çok sayıda zimogen granüllerle dolu olduğu saptandı. Asiner hücresitoplazmasında bulunan krista tipi mitokondriyonların matrikslerinde elektron yoğungranüller tespit edildi. Asinusların açık ve koyu asiner hücrelerden oluştuğu görüldü. Koyuasiner hücreler, açık asiner hücrelere göre serbest ribozom, polizom ve granüllü ER'lardandaha zengin, ER sisternalarının da daha dar olduğu belirlendi. Asinusların lumenlerinde bazenökromatik çekirdekli sentro-asiner hücreler bulunduğu gözlendi. Pankreas loplarınınkendilerine has interkalat, intralobuler, interlobuler ve interlober kanallara sahip olduğusaptandı. Bıldırcınlarda dorsal, ventral ve üçüncü lop ana akıtıcı kanallarının asendensduodeniye açıldığı tespit edildi.Pankreastaki endokrin hücrelerin alfa, beta ve miks adacıklar olarak gruplandığıgörüldü. Alfa adacıklarının A, D, PP ve C hücrelerinden, beta adacıklarının ise B, D ve Ahücrelerinden oluştuğu tespit edildi. Bütün loplarda ara sıra görülen miks adacıkların alfa vebeta adacıklarından oluştuğu görüldü. Alfa adacıkları, dalak lobu ve üçüncü lopta çok sayıdabulunurken, diğer lopların baş kısımlarında az sayıda gözlendi. Beta adacıklarının ise bütünloplarda küçük adacıklar halinde yaygın olarak bulunduğu tespit edildi. Abstract Light and Electron Microscopical Investigations on the Pancreas in Quails (Coturnixcoturnix japonica)The aim of this study was to examine the histological structure of pancreas by light andelectron microscopy in quails. The pancreatic samples taken from twenty quails (healthy and6 monthly of age) were used as materials.In the macroscopic examinations, the quails? pancreas was found to locate between theduodenal arms and had dorsal, ventral, third and splenic lobes. The histological features of theexocrine secretory cells forming the acinus and their relation with the duct system weredescribed. The secretory cells were grouped into acini and were determined pyramidal inshape with basal nuclei, regular arrays of rough endoplasmic reticulum and numerouszymogen granules. Numerous electron-dense granules were determined in the matrix of cristatyped mitochondria which?s found in aciner cell cytoplasm. It was observed that exocrineacinus were consist of dark and light acinar cells. The dark acinar cells were richer in freeribosomes, polisomes, and rough endoplasmic reticulum when compared with the light acinarcells. Besides, rough endoplasmic reticulum cisternae of the these cells were more to getnarrow. Rarely, centro-acinar cells were observed in sections passing through the center of theacini consisting of exocrin pancreas. The morphological features of centro-acinar cells weredetermined to be different from those previously described for acinar cells. All the pancreaticlobes had their own intercalated ducts, intralobular ducts and interlobular ducts. In addition,there were three main ducts (dorsal, ventral and third pancreatic ducts) extending frompancreas to the point where it emptied its contents into the assendens duodenum.The endocrine cells of pancreas were mainly grouped A, B, and Mix islets. The A isletswere determined of which to constitute A, D, PP, and C cells. The B islets were determined ofwhich numerous B cells and islet peripheral to settle from one-two in number of D and Acells. Its observed that the mix islets which are found occasionally in all lobes, are composedof alpha and beta islets. The A islets were numerous in the splenic lobe and third lobe, in theother hand, they were observed a few in the head portion of another lobes. Beta islets weredetermine to be present widespread in small islets in all lobes.Item Domuz timusunda apoptozisin immunohistokimyasal ve ultrastrüktürel yöntemlerle incelenmesi(Sağlık Bilimleri Enstitüsü, 2014) Ceylan, Ahmet; Alabay, Belma; Histoloji-EmbriyolojiItem Ratlarda gebeliğin değişik dönemlerinde trofoblastların özellikleri üzerinde histokimyasal ve immunohistokimyasal çalışmalar(Sağlık Bilimleri Enstitüsü, 2004) Harem, İsmail Şah; Alabay, Belma; Histoloji-Embriyoloji59 SUMMARY Histochemical and Immunohistochemical Investigation on Trophoblasts Characteristic in Different Stages of Pregnancy in Rats In this study, it's aimed to examine trophoblast cells and other placental structures histochemically and immunohistochemically in various periods of gestation in rats. Twenty adult, healty and pregnant rats were used as materials. It was observed that there were significant AB (+) reactions in syncytiotrophoblasts, PAS (+) in giant cells and cytotrophoblasts in the first half of gestation in histochemical examinations. In the second half of gestation, glycogen cells stained PAS/AB (+) coloured in purple granules and small basophilic cells showed gradually increasing AB (+) reactions. In the advanced state of gestations, decidual cells having vacuoles had PAS (+) reactions increasingly. In immunohistochemical examinations, hormonal activities increased significantly in trophoblasts and giant cells during the period staining from the implantation to the end of gestation. Oestrogen and progesterone receptors were found in high density in the cytoplasm and nucleus of the cells. Progesterone receptors decreased slightly towards the end of pregnancy, however, reactions were high in receptors belonging to both hormones.
