Item Seseli gummiferum Pall. ex Sm. ve Seseli corymbosum Boiss. & Heldr. subsp. corymbosum üzerinde farmakognozik araştırmalar(Sağlık Bilimleri Enstitüsü, 2015) Aslan, Sena Seren; Tosun, Alev; EczacılıkThe aim of this study is to perform the phytochemical and biological activity studies on Seseli gummiferum Pall. ex Sm. and Seseli corymbosum Boiss. & Heldr. subsp. corymbosum which are growing in Turkey, from Apiaceae family. Because of the fact that there are limited studies on these species, analysis of the essential oils and biological activity studies have been carried out in this research. First of all, the material of the S. gummiferum and S. corymbosum subsp. corymbosum as our working samples were collected from the nature and authentic samples were deposited in the Herbarium of Ankara University Faculty of Pharmacy. The n-hekzan, ethyl acetate and methanol extracts from the aerial parts and the roots of the each species were prepared. Then, these extracts were examined on thin layer chromatography (TLC). In addition, the essential oils of these species were obtained by hydrodistillation with a Clevenger type apperatus which is regulated according to the European Pharmacopoeia (2010). The composition of the oils were examined on TLC and then analyzed on GC/MS for the comparison of the oils composition. Moreover, the biological activity of the oils have been investigated by the methods of MTT, NO and PGE2, Nf-ĸB and Western Blotting; besides determination of the antimicrobial activity. Regarding to the GC/MS results; in total 29 compounds were characterized representing % 95.1 of the oil of the fruits from S. gummiferum. Germacrene D (% 54.1), sabinene (% 22.4) and bicyclogermacrene (% 5.0) were the major components in SG oil. On the other hand, in total 36 compounds were characterized representing % 93.7 of the fruits oil from S. corymbosum subsp. corymbosum. The main constituents were determined as β-phellandrene (% 29.2), α-pinene (% 9.7), α-phellandrene (% 8.2), germacrene B (% 7.4), bicyclogermacrene (% 6.6), β-pinene (% 4.8) and mircen (% 4.7). In the biological activity studies, the essential oils of the species inhibited production of NO and PGE2, iNOS and COX-2 enzymes, and Nf-ĸB in RAW 264.7 cells stimulated with LPS depend on the concentration. Nevertheless, the screening of the antimicrobial activity of the oils have no satisfied results. Therefore, this study provides important information regarding to use of the oils as a candidate agents against inflammatory disorders and the result related to this activity was debated in this work.
